Why the MICBUSTERS qPCR Quantification Limit Changes by Sample

The useful quantification limit belongs to the complete sample method. It is not a single number printed on the thermocycler. Changing the sample amount, preparation or dilution changes how much original material is represented in a reaction.

Distinguish detection from quantification

The limit of detection describes detection performance under a defined procedure and probability criterion. The limit of quantification describes the lower level at which a quantity can be reported with the method’s accepted performance. A target can be detected without supporting a defensible numerical concentration.

MICBUSTERS technical guidance describes reporting such a detection as +. Use the legend on the current certificate or analyzer sheet for the exact symbol definitions. A plus sign is not zero, and it is not a precise estimate just below the lowest printed number.

What determines the sample-specific limit?

  • The original volume, mass or area represented.
  • The fraction actually carried through preparation.
  • DNA recovery and the approved treatment of the internal-standard result.
  • The extract volume and fraction used in the reaction.
  • Any dilution or cleanup introduced to manage the matrix.
  • The assay’s validated quantitative range and acceptance rules.

Changing one of these factors can change the reporting limit even when the instrument and target assay stay the same. A heavily diluted difficult sample can therefore have a higher reported limit than a clean sample.

Read typical figures as examples

Indicative examples in MICBUSTERS technical guidance
SampleIllustrative LOQHow to use the figure
Typical produced waterApproximately 10¹–10² Gu/mLPlanning example; actual limit follows the individual sample and method
SwabApproximately 10³ Gu per swabNot a value per cm² unless area is known and conversion is justified

These figures are not universal guarantees or instrument sensitivity specifications. The technical guidance also groups solids with swab examples; a solid result needs its own mass or preparation basis and should not inherit a “per swab” unit. Keep the laboratory’s stated meaning of Gu with the result. Do not silently relabel it as viable cells.

A simple dilution example

Suppose a qualified method reports a water LOQ of 20 units/mL. If the extract must then be diluted tenfold, with every other factor unchanged, the equivalent limit becomes 200 units/mL. This is an arithmetic illustration, not a MICBUSTERS performance claim.

Dilution can improve amplification by reducing interference, while reducing the amount of target entering the reaction. A cleaner reaction does not necessarily create a lower limit for the original sample.

How to trend results close to the limit

Keep detections below LOQ separate from quantified values. Retain the reporting limit for each sampling round. A shift from “+” to a quantified result can reflect a real increase, a lower reporting limit or both. A non-detect in a poorly recovered sample is also less informative than a non-detect with acceptable quality and a low method limit.

Where lower limits matter, ask MICBUSTERS about qualified concentration or preparation changes before the next campaign. Collecting a larger representative amount may be more useful than trying to extract certainty from one very low-level reaction.

Frequently asked questions

Does “+” mean the sample is negative?

No. In MICBUSTERS technical guidance it indicates detected target below the quantifiable range. Always use the legend supplied with the current report.

Is Gu/mL a viable cell count?

Do not assume that. Retain the method’s definition and calibration basis; DNA-based quantities do not independently establish viability.
