Using MICBUSTERS Portable qPCR: A Practical Field Guide

A useful on-site qPCR result starts before the thermocycler is switched on. It starts with a defined question, the right sample and a workflow your field team can repeat. This guide brings the practical details together.

Start with the operational decision

Are you comparing an injection-water train before and after treatment, investigating material beneath a deposit, or tracking a coupon over time? Each question changes the sampling plan. A water sample describes the sampled water; it does not automatically describe the biofilm at a corroding surface. Select the location, matrix and reporting unit before ordering the assay panel.

MICBUSTERS combines sampling support, DNA preparation, target assays, portable thermocycling and interpretation. A prepared workflow can typically return targeted information in approximately two hours. Campaign duration also includes access, collection, controls and any repeat extraction. Treat the estimate as planning guidance, rather than a guaranteed turnaround for every matrix.

Build a workflow the team can follow

  1. Define the sample. Record its point, time, volume, mass or surface area and the relevant operating condition.
  2. Preserve or process promptly. Use the procedure qualified for that matrix and downstream test.
  3. Extract with quality evidence. Retain the internal-standard result and any required blanks.
  4. Prepare the run cleanly. Keep sample handling separate from clean reaction setup.
  5. Check the controls before interpreting a number. Review the complete run and the sample-specific quality flags.
  6. Connect the result to the asset. Compare like units and include corrosion and operating evidence.

Keep molecular evidence and corrosion evidence connected

qPCR measures selected DNA targets. It does not directly measure metal loss, prove that every detected organism is alive, or establish that microorganisms caused a particular pit. A positive functional marker identifies genetic potential within assay coverage; expression and activity require additional evidence.

Use the result alongside photographs, deposit observations, chemistry, treatment history and inspection or coupon data. If the microbiology and corrosion evidence disagree, investigate the sampling location and method before increasing treatment on the basis of one number.

Prepare the first campaign with MICBUSTERS

Send us your asset type, intended decision, sample matrices, likely number of samples and working conditions. We can help choose an initial panel and plan training, preservation, controls and reporting. A focused first campaign creates a practical baseline that can be expanded as the team gains experience.

Frequently asked questions

Can a field team use qPCR without a conventional molecular laboratory?

Yes, with training, a suitable clean setup, dependable power and the validated sample workflow. A full laboratory building is not essential, but procedural control remains essential.

Does every sample need a different kit?

Different matrices can require different preparation steps. Agree the workflow for your water, swabs, filters or deposits before collection.
