How to Set Up a Clean Workspace for Portable qPCR

A portable instrument makes on-site measurement possible. A sensible workspace makes the result more trustworthy. Plan the movement of samples, reagents, people and waste before the first tube is opened.

Separate dirty handling from clean setup

Opening deposits and processing swabs can spread sample material onto gloves, tools and surfaces. Preparing reaction tubes requires a cleaner area. Give these tasks distinct places and follow a consistent direction of work. If space is restricted, agree a documented sequence with cleaning and fresh consumables between stages.

Choose a stable surface sheltered from dust, spray and direct sun. A vehicle or portacabin can be practical, provided the location supports the protocol and the site assessment. An exposed tailgate beside active sampling may be convenient for equipment but unsuitable for open reaction setup.

Keep amplified material out of the preparation area

After amplification, reaction tubes contain many copies of the target region. Keep used tubes closed and remove them through the waste route. Opening them beside clean supplies can introduce target DNA that future qPCR reactions detect.

Do not place used tubes, waste bags or dirty cases on the clean setup surface. Keep clean consumables sealed until needed. Use the recommended tips and change gloves after dirty tasks; a glove is only as clean as the last surface it touched.

Use controls to locate contamination

A no-template reaction checks contamination associated with reaction setup and reagents. An extraction blank follows preparation. A field-handling blank can reveal contamination introduced during collection. Choose the set that matches your workflow rather than assuming one blank monitors every stage.

If a blank is positive, retain its target identity and curve information. Review the approved rule for the affected batch. A low positive sample near a contaminated blank needs a different response from a cleanly controlled strong positive. Do not subtract a blank value and declare the problem solved without a validated method.

A practical setup sequence

  1. Select the clean and sample-handling areas before unpacking.
  2. Prepare the surfaces and tools using the approved cleaning procedure.
  3. Set out labels and the run plan so open-tube time stays short.
  4. Handle samples in their designated area and keep the clean supplies separate.
  5. Prepare reactions in the clean area with fresh handling precautions.
  6. Close tubes before moving to measurement and leave used tubes closed.
  7. Record the control results and any deviations in the campaign log.

Do not improvise the decontamination chemistry

The cleaning procedure must suit the instrument and surfaces. Keep incompatible cleaners and industrial sample residues apart, and follow the manufacturer instructions. Avoid introducing moisture or cleaning chemicals into the instrument’s optical or electrical components.

Internal-standard recovery can be acceptable in a contaminated sample. That is why sample-quality monitoring and contamination control remain separate parts of the plan.

Make the layout part of training

MICBUSTERS can help your team rehearse the full sequence using the space and equipment available at the site. Include where the operator puts a used pipette, changes gloves and discards closed reaction tubes. These small decisions reduce avoidable handling errors during a busy sampling campaign.

Frequently asked questions

Do I need a laboratory building?

Not necessarily. You do need an appropriate controlled setup and a workflow that keeps sample material and amplified DNA away from clean reaction preparation.

Can I open completed reaction tubes to inspect them?

Keep them closed in the routine workflow. Opening amplified tubes can contaminate later preparations.
