Using Portable qPCR in Hot and Humid Field Conditions

A thermocycler controls reaction temperatures during the program. It does not protect reagents left in a hot vehicle before the run. Plan storage, transport and final reaction setup as separate parts of the field workflow.

Separate storage conditions from operating conditions

The temperature allowed for long-term reagent storage can differ from the allowed transport excursion or instrument operating range. A compact case makes transport convenient, but it is not automatically insulated or temperature controlled. Review the label and current instructions for every component in the campaign.

Target tubes, DNA preparation reagents, preserved samples and retained extracts can each have different requirements. Do not apply the preservative’s permitted holding condition to all other materials in the case.

Protect the target tubes before setup

MICBUSTERS’ field checklist flags heat exposure, particularly above 30°C, as a concern for extraction materials and target tubes. This is a prompt for careful planning, not a universal hard limit for every component or an instrument operating specification. Use current product-specific instructions for acceptance of an excursion.

Store packs as directed, keep them sealed against moisture and remove only what is needed for the session. Prepare the final target-tube steps in a room with controlled moderate temperature and humidity where practical. Keep tubes away from direct sun, hot metal surfaces and open doors exposed to spray or dust.

Allow controlled acclimatization

Cold equipment or packs entering warm humid air can collect condensation. Allow suitable acclimatization according to the instructions, keeping packaging closed where appropriate. Do not open moisture-sensitive material simply to make it warm faster.

There is no fixed warm-up time that applies to every arrival condition. Consider the temperature difference, condensation risk and instrument instructions. A stable control result after proper setup is more useful than assuming a short elapsed time guarantees readiness.

Prepare for the whole working day

  • Identify the storage location before leaving the office.
  • Check whether the site refrigerator or approved transport arrangement is available.
  • Limit time in parked vehicles and on exposed decks.
  • Track excursions where required by the handling procedure.
  • Set out only the material needed for the next preparation.
  • Keep labels, lot numbers and expiry information with the run record.

If a shipment or case has been overheated, isolate the affected material and ask for a disposition using the documented exposure. A normal-looking tube does not establish reagent performance. Do not hide an excursion by mixing the affected stock into a new pack.

Include operator comfort in the workflow

Heat, cramped space and poor lighting can make careful pipetting and recordkeeping harder. A sheltered preparation area can improve both working conditions and consistency. Plan breaks and sufficient bench space alongside the analytical supplies.

MICBUSTERS can help assess a proposed offshore, tropical or remote-site workflow. Share the expected temperatures, humidity, travel time and available facilities so the handling plan fits the actual campaign.

Frequently asked questions

Can I leave the qPCR case in a parked car?

Do not assume that it meets reagent storage conditions. Parked vehicles can create substantial heat exposure; use the component-specific handling plan.

Is there a universal stabilization time after switching on?

No. Follow the current instrument instructions and account for arrival temperature and condensation, rather than using a fixed time for every environment.
